Stroke modelling in sheep: methods, applications and contributions to translational research

Synopsis:
Although indispensable for basic research, rodent stroke models offer limited comparability with human stroke. This is believed to hamper the transfer of promising novel therapeutic and diagnostic procedures for stroke from the lab to the clinics. Large animal models could fill this translational gap, verifying the impact of such procedures in a gyrencephalic brain and in an experimental setup closer to human stroke care. While being laborious to employ, large animal models offer a number of additional advantages such as compatibility with clinical imaging technology.
The talk will briefly review existing large animal stroke models and will provide details on stroke modelling in sheep. It will also provide examples on how the model has been used in translational stroke research spanning from a cell therapeutic approach, over an innovative approach to preserve the penumbra to a direct comparison between PET and MRI-based penumbra assessment in humans and sheep.
