Title: “The trafficking of membrane proteins to the cell surface”
Abstract: Secreted proteins underpin myriad physiological processes, ranging from immunity and inter-cellular signalling to dynamic remodelling of the extracellular matrix. After biosynthesis and post-translational modification, these cargoes must traverse the Golgi apparatus, where they are selectively sorted into carriers bound for the plasma membrane or the extracellular milieu. The molecular logic governing this journey remains incompletely defined.
Here, we integrate super-resolution microscopy with transient CRISPR-mediated knock-outs, quantitative pulse-chase trafficking assays and designer fluorescent cargo constructs to dissect the spatiotemporal rules of Golgi exit. Our data uncover novel machinery and mechanisms for Golgi export. Collectively, these findings refine the conceptual framework of secretory-pathway organisation and provide a predictive toolkit for manipulating protein secretion in health and disease.
