Title: "Autotransporter adhesins in Gram-negative bacteria: structure, function, evolution"
Summary: Autotransporters - or Type V secretion systems - are simple secretion systems that translocate surface proteins across the outer membrane of Gram-negative bacteria. Mostly, these proteins then remain anchored in the membrane and fulfil key functions in pathogenesis. Some of the major adhesins of Gram-negative human pathogens are part of this large family, mediating attachment to host cells and/or initiating biofilm formation.
Over the last years, my group has been working on both the translocation mechanism across the membrane, and the binding functions of select type-V-secreted adhesins. We use a broad range of methods from infection biology to bioinformatics and NMR. Our model systems include both human and animal pathogens - we try to learn from the similarities and differences of their adhesin virulence factors, aiming to understand what defines host specificity, and how adhesion could be used as a target for new antibacterial drugs.
