Abstract: Collagen IV alpha 1 (COL4A1) and alpha 2 (COL4A2) are primordial extracellular matrix proteins that are core components of basement membranes in all organs. COL4A1 and COL4A2 mutations cause a multi-system disorder for which cerebral small vessel disease is a major consequence. We use genetic approaches to understand the mechanisms by which COL4A1 and COL4A2 cause defects during developmental angiogenesis and age-related cerebrovascular dysfunction.
