Abstract: The role of tissue stiffness in controlling cell behaviours is well established. This has been shown across cell types and includes behaviours such as cell differentiation and even drug susceptibility, with clear implications for health and disease. In experimental investigation of mechanosensation the use of bioengineered gels with defined mechanical properties have become essential to mimic varying tissue microenvironments. However, I show here that the stiffness experienced by the cell is not only determined by the actual stiffness but rather incorporates the interplay between contractility, cell adhesion and mechanical response. I will also discuss how these observations scale up in whole tissues looking at recent experimental observations in epithelial sheets, and at the role geometrical constraints play in three-dimensional tissues.
